Just my personal opinion. Why shoot a 250 gr. .45 when a .44 does it so much better. For the same weight the .44 will penetrate better. For MY .45 Colt loads I see little gain until I go 300 gr. Not that I blow the roof off with hot loads. My favorite is a case full of AA1680 under a 300 gr. LEE FNGC that gives about 1000 fps. I like 4227 too but it is real dirty unless I use magnum primers. The old Unique load from 8-10 grs. is a nice stand by.
I've sized down .458 300 gr. JHP but the jackets seem real thick as their original intent might suggest. They do shoot well though. LEE also makes a 340 gr. mould for the .45-70 that I re-size to .452 and it makes a fun heavy. I have several .45 Colts but I've just never really jumped on the band wagon. In a carbine the thin rim has caused me extraction issues in the past. I just tend to gravitate the the .44 more. There is not a whole lot of difference when using the same weight bullets except you'd figure a .43 would have a superior ballistic coefficient than a .45. On paper it does anyway. The argument will live on. More fun to have both anyway. There will always be a macho, knuckle dragging element, of one-up-man-ship having a .45 over a .44 (that's stretching it too as it's really a .43). There is a bit of meat to the idea that the .454, perhaps even the .45 ACP) has rendered the .45 Colt somewhat obsolete except for a light weight trail gun of historic providence and flair.
*You guys know I'm playing the devils advocate for discussion.

For a little fun, I'm not a huge fan. The .44 is superior.
