As others said, GP100 is cheaper, etc.

But, the new Python definitely can. You could also go 10mm mag.

I'm doing something similar. I had reamers made for a rimmed straight-wall .40 caliber magnum cartridge. Similar to the old .401 Powermag, but in true .40/10mm. Essentially it will be a 10mm mag, but rimmed.

Some would likely argue, "why not go .41 mag?" These days, with the popularity of 10mm, bullet selection is better for 10mm than .41. Even though some factory ammo is available for .41 it's really a handloader's cartridge.
