Some of the big Nitro Express cartridges didn't do so well. It appears that the larger bullets didn't penetrate so well due to larger diameter.

I'm not sure how that carries over into revolver rounds. Surely, there is some crossover with the various 475 and 500 calibers.

But, consider this. There is .035" difference between the two bullet diameters. The difference between the 44s and 45s is only .023" different. Since those two have survived well over 100 years, surely we have room for the 475s and 50s.
