I personally prefer a flatter point cast bullet. I used a .510 GNR revolver and 410 gr. bullet at 1100 fps. I was really torn about the place to shoot. I didn't feel like I could hit that neck bone. I was afraid I'd shoot it's goozle, then the chase would be on. The guide told me to shoot it on the point of the shoulder. I did, it folded on it's nose, but still very much alive. Double lunged, with huge lungs, but they can't stand on no front legs. The bullet, even going that speed broke both shoulders and exited. I finally walked up and it took a close up neck shot to instantly end it.
It was drizzling and I shot from about 25 yds. off hand. I hit within an inch or two of where I was aiming. John Ramsey Miller shot a smaller bison at the same time with a .41 GNR. The 240 gr. bullets passed through the lungs and threw up water 50 yds. distant. In an out like warm butter, didn't look like they even slowed down that much. (I had the same experience with the .510 and a poorly hit 80 lb. sheep!). Onlookers thought he shot over. He hit his mark, just no bone. We chased a while, maybe 5-6 shots total. Not what a sportsman desires, humbling, and a good lesson. Sometimes it just goes that way no matter how much you prepare.

Sounds like you have it figured out to prep. Any good .41/.44 load with a tough bullet in the sweet shot will do it. A miss is a miss, no matter the caliber.
